I am trying to help a friend with her iPhone. She recently purchased an iPhone 6SE. We were trying to lock her screen orientation to "Portrait," by going to the Control Center, but the option wasn't there. Does anyone know if this has recently been changed? If so, how do we fix it?

I just checked and it has a lock orientation switch on the first page under do not disturb. The second page has other stuff, so maybe check both pages if you haven't already.

Yeah, it's there. like prev poster said, it's on the first default page in the control center. I personally use this all the time, as I hate it when my iPhone 6S plus goes into landscape mode unless I'm playing certain games that automatically get landscape mode

Hi! What page were you on in the control center? In ios. 10, Apple changed to a 2-page control center. go to the first page and swipe until you hear lock orientation and toggle it on. Just make sure the device is in the orientation you'd like it to be before doing it. Anytime she needs to change it, she'd just go back to that same page and switch it to off.
The 1st control center page has things like your camera, air drop, nightshift and orientation locking on it. The 2nd page is dedicated to your music or other music apps that can be controled from control center. Good luck and hope this helps!
