I have a concert that am using my Bluetooth headphones to listen to my VOice over,
but when i go away from my laptop, it disconnects, and when i back again my laptop is connected to external speakers and VOice over will speak from these speakers, so, is there a hot key that we can assign to change audio output without VOice over or mouse?
Need it urgent please because my concert is tomorrow.

AppleScript?
I've previously used An AppleScript in combination with Keyboard Commander to change sound output device.

toothfairy app
There is a mac app called toothfairy, that does exactly this. Its fully accessible, just add the device you want to the list, set a hot key for it and your done. The hotkey both disconnects and reconnects the output. For instance, whenever I press control shift b, the app connects to my bose headphones, and when I press it again it disconnects and the audio reverts back to the speakers.

I installed the app, seems very accessible and i set my short cut, but should my bluetooth still connected? i mean if i go away and came after 30 minutes, wafter i press this shortcut, will it connects or i should connect my Bluetooth device again?

Depends on the device
It depends on a lot of factors, such as if the device has a, shut off after x amount of time if no audio is playing etc, if you have the device with you and walk away to the point where your out of range and the device disconnects itself... Generally speaking though, assuming the device doesn't have a disconnect after x time of beeing idle with no audio playing yes, it should in theory, stay connected.
Voice over output changed
Something strange happened now is, when i connect a jack to the audio output, Voiceover output changed to the headphone output, do not know why is this,
and can i ask Voice over for not doing this even if i connect A headphone jack?

I installed the app, and i set a shortcut, but it does not cycle between speaker output and my bluetooth speakers, just, am i missing something?

It should cicle between the two. Did you add the device to toothfairy? I've never had this problem, for me it always works. Also, as far as the headphones... That's possible, but you'd need to use audio hijack, but its normal behavior that voiceover or the audio, switches automatically to the headphones via the jack, but yeah, that can be well not per say stopped, but with audio hijack you can kind of make it happen.
